PMMA bone cement containing long releasing silica-based chlorhexidine nanocarriers

چکیده
Prosthetic joint infections (PJI) are still an extremely concerning eventuality after replacement surgery; growing antibiotic resistance is also limiting the prophylactic and treatment options. Chlorhexidine (a widely used topical non-antibiotic antimicrobial compound) coatings on silica nanoparticles capable of prolonged drug release have been successfully developed characterised. Such nanocarriers were incorporated into commercial formulation PMMA bone cement (Cemex), without adversely affecting mechanical performance. Moreover, containing showed superior activity against different bacterial species encountered in PJI, including clinical isolates already resistant to gentamicin. Cytocompatibility tests non inferior performance cements chlorhexidine releasing equivalent formulation.

Bisphosphonates in bone cement inhibit PMMA particle induced bone resorption.
OBJECTIVE Wear particle induced bone resorption is thought to be one of the mechanisms that contribute to implant loosening. It has previously been shown that macrophages, in response to polymethylmethacrylate (PMMA) particles, differentiate into bone resorbing osteoclasts, and that this process is inhibited by a bisphosphonate, etidronate (EHDP). The aim of this study was to determine whether ...
متن کاملEffects of Incorporating Carboxymethyl Chitosan into PMMA Bone Cement Containing Methotrexate
Treatment of bone metastases usually includes surgical resection with local filling of methotrexate (MTX) in polymethyl methacrylate (PMMA) cement. We investigated whether incorporating carboxymethyl chitosan (CMCS) in MTX-PMMA cement might overcome disadvantages associated with MTX. To determine the optimal CMCS+MTX concentration to suppress the viability of cancer cells, an integrated microfl...

INTRODUCTION Polymethyl methacrylate (PMMA) beads containing gentamicin are still an effective treatment for osteomyelitis. However, the emergence of anti-microbial resistance in general and the increasing prevalence of MRSA in nosocomial infections indicate the need for new antibiotics. Anti-microbial peptides (AMP) are a new promising class of antibiotics and have a low tendency to induce ant...

Poly (methyl methacrylate) (PMMA) bone cement is widely used in vertebral body augmentation procedures such as vertebroplasty and balloon kyphoplasty. Filling high modulus PMMA increases the modulus of filled verterbra, increasing the risk of fracture in the adjacent vertebra. On the other hand, in porous PMMA bone cements, wear particle generation and deterioration of mechanical performance ar...
متن کاملPmma Bone Cement: What Is the Role of Local Antibiotics
Due to its primary fixation purpose antibiotic loaded bone cement (ALBC) has been classified as a class III medical device according to the CE standard for the fixation of primary or revision implants. Because of the high initial release of local antibiotics into the joint cavity, ALBC has also been successfully used for decades for prophylactic reasons aimed at prevention of bacterial colonisa...
